Objective To explore the regulatory mechanism of m6A in Parkinson's disease cell model. Methods The content of m6A in the neurons of Parkinson's disease (PD) induced by 6-OHDA was observed, and the expression levels of methyltransferase and demethylase which regulate m6A were detected. After FTO knockdown, the expression of PD disease related genes and dopamine signaling pathway related genes were observed. Results FTO was significantly increased in PD model cells, and the relative content of m6A decreased to some extent. In FTO knockdown cells, the expression of genes closely related to PD did not change significantly, but the expression of GRIN1, a dopamine signaling pathway related gene, was significantly decreased. Conclusion The effect and mechanism of FTO on dopamine metabolism in Parkinson's cell model were revealed.
