Abstract
[Objective]To investigate the damage effects of bisphenol A (BPA) on human embryo-liver L-02 cell line and the mechanisms of it. [Methods] Human embryo liver L-02 cells were exposed to different concentrations of bisphenol A (1, 10, 50, 100 mu mol/L) Survival rate, level of superoxide dismutase (SOD), glutamylcysteinylglycine (GSH), catalase (CAT), Malonyldiadehyde (MDA), apoptosis rate and the expression level of p53, caspase3 mRNA were measured. [Results] After exposure to bisphenol A, the survival rate of L-02 cell was proportional to the concentration of bisphenol A. Meanwhile, SOD level decreased significantly, GSH, MDA levels decreased at 1 mu mol/L of bisphenol A and increased as the concentration of bisphenol A increased.respectively. CAT level increcised below 10 mu mol/L of bisphenol A then increased significantly as the concentration of bisphenol A increased. Apoptosis and expression levels of p53, caspase3 mRNA increased significantly at 50 and 100 mu mol/L bisphenol A respectively. [Conclusion] Bisphenol A: has cytotoxic effect on and oxidative damage to L-02 cell line, and induces apoptosi.s of L-02 cell line. Oxidative damage may be orie of the mechanisms of apoptosis in L-02 cell line induced by bisphenol A.
