•A simplified and validated method for allicin analysis in garlic was developed.•An optimal solution and sample preparation for allicin extraction was established.•Highest allicin was extracted from fresh garlic, followed by freeze-dried cloves.•The stability of allicin extract under different storage conditions was determined.•Allicin concentration was accurately determined in eight garlic varieties.
