Abstract
Our aim was to study the effect of the ethanol and hydrogen peroxide on EGFR expression and their phosphorylation at Tyr845 in the hepatic cells (WRL68). For the study, we used the WRL68 cells cultured in MEM supplemented with 10% fetal bovine serum, 2mol/L glutamine, 1% non‐essential amino acid. Cells were kept at 37°C, under an atmosphere of 5% CO2. The cells were treated with ethanol at concentrations of 5, 12, 25, 50, 100, 200, 400 and 800 mg/dL, and hydrogen peroxide at concentrations of 0.34, 3.4, 34, 340, 3400 ug/dL. The EGFR expression was analyzed by WB and the phosphorylation was detected by IP and WB. Ethanol and hydrogen peroxide did not modify EGFR expression at 24 h to exposure. However ethanol decreased the EGFR expression (20–50%) before to 24h to exposure and phosphorylation at Tyr845 (40%). While hydrogen peroxide induced changes in EGFR expression (10–50% less) and phosphorylation at Tyr845 was inhibited (50%). In conclusion, ethanol and hydrogen peroxide diminish EGFR expression and phosphorylation at Tyr845 in hepatic cells WRL‐68.
