The Genetic Toxicity Of Fumonisin B1 On Liver Cell

Fumonisin B1 is a mycotoxin produced by Fusarium verticillioides, frequently associated with corn. It produces species-specific and organ-specific toxicity, including equine leukoencephalo -malacia, porcine pulmonary edema, and hepatic or renal damage in most animal species. Fumonisin B1 perturbs sphingolipid metabolism by inhibiting ceramide synthase. FB1 is mutagenic in cell cultures, but the mechanism of its genotoxicity is not understood. We exposed HHL-16 cell to FB1 with the aim of finding the mechanism of how FB1 affect liver cell from a genetic standpoint. Our results indicate that FB1 treatment (5uM-80uM) did diminish cell viability, but cell viability was not reduced obviously by FB1 (100uM and 500uM). The level of intracellular ROM was increased in a dose dependent manner associated with an increase in FB1. The dose -dependent effect between the concentration of FB1 and DNA damage was observed by the comet assay. DNA and RNA variation is shown that FB1 maybe affect the genetic transcript.
