Association Of Ppar Gamma 2 Gene Pro12ala Allelic Variant With Obesity And Metabolic Syndrome Components In A Population Of Maracaibo

Abstract
Introduction: In this study the association of rs1801282 allelic variant, g. 68777C G (Pro12Ala) of PPAR gamma 2 gene with anthropometric and metabolic abnormalities was evaluated in a cohort of individuals with Metabolic Syndrome (MS) in Maracaibo Municipality.Methodology: The sample consisted of 127 individuals (81 with a diagnosis of MS, according to standardized criteria of expert panels IDF / AHA / NHLBI and 46 healthy subjects). Polymorphic versions of PPAR gamma 2 gene were analyzed by PCR-RFLP.Results: The frequency of the polymorphism of PPAR gamma 2 gene in the study population was 89 % for the Pro12Pro homozygous genotype and 11 % for Pro12Ala heterozygous genotype. The Ala12Ala homozygous variant genotype was not detected. The Pro12Ala heterozygous genotype was observed more frequently in patients in the MS group, although no significant genotypic (p=0.528) or allelic (p = 0.602) frequencies differences between SM and control groups was found. Pro12Ala genotype exhibited a higher average value in percentage of body fat, waist circumference and HDL-C, as well as lower values of blood pressure, total cholesterol, triglycerides, LDL-C and VLDLC, however there was no statistical difference. Higher average value of BMI in PPAR gamma 2 Pro12Ala genotype carriers (p=0.038) was observed. Nevertheless this polymorphism had no greater effect on the metabolic profile in these individuals with high BMI. 42.9% of the individuals carrying the Pro12Ala genotype and 30% of Pro12Pro carriers reported a family history of obesity, which may indicate a contribution or interaction of other genetic components.Conclusions: The results of this study suggest the association of Pro12Ala genotype with risk of obesity and lack of association between polymorphism of the PPAR gamma 2 gene with metabolic syndrome, with only modest effects on individual components
