Abstract
The anti-inflammatory activities of AD-3, 15-p-chlorobenzylidene-14-deoxy-11, 12-didehydroandrographolide, which was synthesized from andrographolide (AD), was investigated by models of ear edema induced by dimethylbenzene in mice and foot edema induced by albumn in rats. Compared with AD, the mother compound, the mechanisms of enhanced anti-inflammation of AD-3 were preliminarily clarified. It was found that AD-3 (1.35mmol/kg) showed stronger anti-inflammation activity than AD on ear edema induced by dimethylbenzene in mice (1.35mmol/kg, P<0.01). The results about the effects of AD and AD-3 on NO and PGE(2), the inflammatory mediators, suggest that both of AD-3 and of AD have anti-inflammatory activities based on the inhibition on NO. Both of AD-3 and AD played anti-inflammation activities by inhibiting PGE(2), and AD-3 showed stronger inhibitory activity on PGE(2) than AD.
