Abstract
Objective: To explore the characteristics of DNA fingerprint in the hearts of Martes zibellina L. and set up a specific molecular biology method to identify Martes zibellina L. hearts from its counterfeits hearts. Methods: Mitochondrial DNA was extracted from fresh heart of Martes zibellina L. and spurious animal tissues by alkaline lysis method. The specific fragment was amplified with PCR by designing specific primers and the PCR products were sequenced and blasted in Genbank. Results: Complete 16.8 kb mtDNA was separated from fresh heart of Martes zibellina L. and spurious animal tissues of chicken hearts, duck hearts, goose hearts and rabbit hearts. However only the fragments with 310 bp can be amplified by PCR as a template of Martes zibellina L. Moreover, PCR products sequence revealed that the 310 bp segments were identical to that described in GenBank accession number AB026109.1. Conclusion: The Mitochondrial DNA from the heart of Martes zibellina L. and its counterfeits possesses the same fingerprint feature, and the fragments of 310 bp amplified with PCR can be used for specific marker in identifying Martes zibellina L. heart from spurious hearts.
