•Buspirone and zolmitriptan synergistically reduce L-DOPA-induced dyskinesia in a rodent Parkinson's disease model.•The buspirone and zolmitriptan combination is safe and does not lead to tolerance development.•The anti-dyskinetic effect of buspirone and zolmitriptan combined is superior to amantadine.
