•Pancreatic ductal adenocarcinoma cells primarily employ integrin α6β4 to adhere to their ECM.•Integrin α6β4 is assembled into hemidesmosomes in culture that are remarkably complete in composition and structure.•Depletion of integrin β4 blocks the proliferation of pancreatic ductal adenocarcinoma cells.
