•Intron retention (IR) is a new pre-symptomatic marker of aging.•The pre-symptomatic state in the liver in terms of gene expression was recovered to the healthy state by Japanese herbal medicine.•The IR accumulated by aging stress was recovered to the healthy state by Japanese herbal medicine.•The pre-symptomatic (pre-disease) state is reversal to the healthy state by Japanese herbal medicine.
