•PPI G4 dendrimer binds about 30 molecules of tegafur (TF).•Complexation of PPI G4 with TF is exothermic and occurs spontaneously.•TF molecules are bound with surface part of PPI G4 macromolecules.•Hydrophobic tetrahydrofuran part of TF stabilizes supramolecular complex.
