•Out-of-use wool was converted to a modifier for the electrochemical sensing platform.•The Pd2+ ions were adsorbed onto reduced-keratin (KerSH) extracted from waste wool.•KerSH-PdNPs were co-electrodeposited onto the GCE surface by chronoamperometry.•The dsDNA adsorbed onto GCE/KerSH-PdNPs by self-assembly of KerSH's NH2 groups.
