Abstract
探讨人参皂苷Rg1对H9c2心肌细胞缺氧/复氧(hypoxia/reoxygenation,H/R)损伤的保护作用及其信号通路.建立H9c2心肌细胞的H/R模型,分成不同的处理组,利用CCK-8(cell counting kit-8)法检测心肌细胞的活性,Brdu法检测H9c2细胞的增殖,检测心肌细胞caspase-3的活性,Western blot法检测蛋白表达.利用流式细胞仪检测心肌细胞凋亡水平.人参皂苷Rg1可抑制H/R导致的心肌细胞凋亡和caspase-3的活性;可促进核因子红细胞2相关因子2(nuclear factor erythroid-2 related factor 2,Nrf2)核转录,同时可增强下游基因血红素加氧酶1(heme oxygenase-1,HO-1)表达;可随着浓度的增加(10,20,40,60 μmol·L-1)Nrf2核转录和HO-1表达增加;然而,心肌细胞转染Nrf2-siRNA后人参皂苷Rg1保护心肌细胞的作用明显减弱.人参皂苷Rg1可通过激活Nrf2/HO-1信号通路保护心肌细胞.
