Abstract
•Conk-C3 is a novel Kunitz-type K+ channel blocker isolated from cone venom.•MD simulations suggest that Conk-C3 traps K+ ions in cryptic coordination sites.•This prediction is supported by mutagenesis and by electrophysiological assays.•The results provide a molecular description of K+ channel “lid-block” by an animal toxin.
