•HSPBP1 interacted with RIG-I and the interaction was enhanced upon virus infection.•HSPBP1 is a positive regulator of RLR signaling.•Knockdown or knockout of HSPBP1 inhibits RLR signaling.•HSPBP1 inhibits RIG-I K48-linked polyubiquitination to stabilize it and strengthen its cellular antiviral response.
