重组酶聚合酶扩增技术检测霍乱弧菌

Abstract
本研究根据编码霍乱弧菌外膜蛋白W(ompW﹚的基因保守序列,设计重组酶聚合酶扩增(RPA﹚技术特异性引物,建立ompW基因的RPA检测方法并测试其特异性、灵敏度和应用效果.结果显示,建立的RPA检测方法特异性好,能够从霍乱弧菌中检测到280 bp的特异性条带.对比RPA和实时荧光聚合酶链式反应(PCR﹚,灵敏度一致,均达到0.1 ng/μL,说明所建立的RPA方法具有较好的灵敏度.综上所述,本研究建立的RPA方法检测ompW灵敏度高,特异性强,无需特殊的仪器及设备,适合实验室快速检测ompW.
