毕赤酵母工程菌产乳糖酶发酵条件优化的研究

Abstract
目的 优化毕赤酵母工程菌株VGal3186的发酵条件.方法 对发酵培养基中的碳源、氮源、金属离子和pH、温度、接种量等发酵条件分别进行了单因素和正交试验优化,并在50 L发酵罐中验证.还对所产重组米曲霉乳糖酶进行酶学性质研究.结果 优化后的发酵培养基为葡萄糖45g/L、甘油15g/L、蛋白胨20 g/L、酵母提取物20g/L、KH2PO4 4 g/L、MgSO4 9 g/L、CaC12 0.5 g/L、FeSO490 mg/L、CuSO4 6mg/L、K2SO4 18.2g/L,发酵条件为pH 7.0、发酵温度30℃、接种量7%,转速300 r/min.摇瓶发酵酶活性可达689 U/mL,较优化前提高了35.2%.50 L发酵罐中酶活性可达5264 U/mL,酶学性质表现更好.结论 毕赤酵母工程菌株VGal3186经发酵优化后产量更高,产乳糖酶酶学性质好,在食品工业中应用潜力巨大.
