Abstract
目的:探讨Ras癌基因诱导的miR-182-96-183高表达机制.方法:采取H-ras12V转基因小鼠肝癌及癌旁组织,miRNA高通量测序检测miR-182-96-183基因表达水平.Westernblot检测调控miR-182-96-183表达的相关信号通路活化状态及其上游转录因子蛋白表达水平.结果:miRNA高通量测序结果表明:与PT相比,T中miR-182-96-183 mRNA表达显著升高(P<0.01).Westernblot检测结果表明:与PT相比,T中ERK、mTOR活化水平显著升高,SREBP-1c蛋白水平显著升高.结论:Ras癌基因可能通过活化mTOR通路激活SREBP-1c上调miR-182-96-183的表达水平.
