百合知母汤对CUMS抑郁症大鼠海马中ERK1/2信号通路关键分子的影响

Abstract
目的:研究百合知母汤对CUMS抑郁症大鼠ERK1n信号通路中的关键分子的影响.方法:SPF级SD大鼠50只,随机分为空白组、氟西汀组、百合知母汤中剂量组、百合知母汤大剂量组、模型组,每组10只.除空白组外,其余大鼠CUMS法制造抑郁症大鼠模型,共计21 d;造模完毕后,各组分别给予氟西汀(1.8 mg/kg)、百合知母汤(1.5 mg/kg)、百合知母汤(3.0 mg/kg),空白组、模型组均给予等体积生理盐水,共计28 d;给药完毕后,取大鼠海马组织,分别用Western-blot法、qPCR法、免疫组化法检测PKC蛋白、MEKmRNA、ERK1/2蛋白的表达量.结果:与模型组比较,百合知母汤中、高剂量组海马组织中的PKC蛋白、MEKmRNA、ERK1/2蛋白的表达量均升高(P<0.05或P<0.01).结论:百合知母汤的抗抑郁机制可能与上调CUMS抑郁症大鼠海马组织中ERK1/2信号通路的关键分子PKC蛋白、MEKmRNA及ERK1/2蛋白有关.
