地五养肝胶囊对肝纤维化大鼠肝脏Hedgehog通路的调节作用

Abstract
目的:探讨地五养肝胶囊(DWYG)对肝纤维化大鼠肝脏Hedgehog(Hh)信号通路的影响.方法:Wistar大鼠随机分为空白对照组、肝纤维化模型组和DWYG干预组,四氯化碳(CCl4)诱导肝纤维化模型.应用HE和Masson染色检测不同处理组大鼠肝组织损伤和纤维化程度;应用Western Blot和Real-time qRT-PCR分别检测不同处理组大鼠肝组织Hh信号通路的蛋白和基因表达水平.结果:与空白对照组相比,肝纤维化模型组大鼠出现明显肝组织损伤和纤维化改变,且纤维化肝组织中Shh、Gli1、Smo和Ptc的蛋白和基因表达水平显著上调(P<0.05);而与肝纤维化模型组相比,DWYG干预可显著减轻肝组织损伤和纤维化程度,同时显著下调纤维化肝组织中Shh、Gli1、Smo和Ptc的蛋白和基因表达水平(P<0.05).结论:DWYG抗纤维化效应与其抑制纤维化大鼠肝组织中过度活化的Hh信号通路相关.
