大孔吸附树脂精制黄芪总皂苷工艺研究

Abstract
目的:系统研究影响大孔吸附树脂吸附分离的因素,筛选黄芪总皂苷的最佳吸附树脂和最佳工艺.方法:根据树脂的吸附-洗脱性能、吸附动力学曲线,从8种树脂中筛选出最佳吸附树脂,以黄芪总皂苷提取率、纯度为综合评价指标进行工艺优化.结果:AB-8、XDA-5、XDA-1三种树脂具有较优的吸附-洗脱性能,并依据吸附动力学曲线,确定AB-8型为最佳吸附树脂.工艺优化结果为AB-8型树脂,精密吸取样品液0.876 mg·mL-1(总皂苷与干树脂质量比为34.6 mg·g1)100 mL上柱,蒸馏水洗至无还原糖反应后,依次用10 BV 0.2%~0.3%NaOH溶液、10%乙醇和70%乙醇进行洗脱,流速为1~3 mL·min-1,收集70%乙醇洗脱部分,所得产物中总皂苷提取率和纯度分别为47.79%、68.98%.结论:该工艺简便可行,可较好的精制黄芪总皂苷.
