桑葚多糖T3-3分离及秀丽隐杆线虫抗衰老活性研究

Abstract
[目的]研究桑葚多糖T2提取工艺优化,并对其抗衰老活性进行分析.[方法]以秀丽隐杆线虫为模型,用寿命、移动力分析、吞咽频率、产卵量和肠脂褐质综合评价不同剂量范围的桑葚多糖(T3-3)抗衰老活性.通过正交和优化实验确定最优提取工艺条件为:提取温度100℃,提取时间2 h,提取次数3次,料液比1:30,提取率最高达8.48%.[结果]抗衰老活性研究表明,150 mg/L的T3-3能增强秀丽隐杆线虫的抗衰老能力.[结论]该提取工艺1)延长了秀丽隐杆线虫的寿命,秀丽隐杆线虫平均寿命延长了10.38%(P<0.05),最大寿命延长了28.57%(P<0.05),50%存活率对应天数延长了0.91%.2)秀丽隐杆线虫吞咽频率增加,吞咽频率比对照组高18.78%.3)产卵量无统计学差异.4)减少秀丽隐杆线虫肠脂褐质的产生量,比对照组平均荧光强度低11.14%(P<0.05),实验末期,对照组的平均荧光强度增大了12.47%.
