Abstract
目的:探讨牛膝治疗膝骨关节炎的有效活性成分及作用机制。方法:检索TCMSP中牛膝含有的化合物,筛选其活性成分,进而获取活性成分的潜在靶点;通过GeneCards数据库挖掘膝骨关节炎靶点;结合化合物及疾病靶点,构建化合物-靶点网络,进行GO富集及KEGG通路富集分析。结果:牛膝含有23个活性成分,对应膝骨关节炎136个靶点;获得1 592条GO生物过程相关条目与19条信号通路,涉及IL-17信号通路、TNF信号通路、凋亡信号通路、PI3K/AKT信号通路等通路。结论:牛膝主要通过调控膝关节内免疫炎症反应减轻炎性损伤,并抑制软骨细胞凋亡,保护软骨。
