EB病毒mir-BART-5促鼠成纤维细胞恶性表征机制研究

Abstract
目的 探讨EB病毒mir-BART5可促进鼠成纤维细胞恶性表征的作用机制. 方法 生物信息学分析显示miR BART5与miR-18序列相近,miR 18的靶基因表达的结缔组织生长因子(CTGF)可促进细胞增殖.在鼠成纤维细胞NIH/3T3中瞬时转染mir-BART5-mimics使其表达上调,采用Western blot检测对照组与转染组细胞的CTGF表达,采用体外软琼脂集落生成试验观察两组细胞生长的恶性表征. 结果 转染组细胞在软琼脂上呈恶性表征.CTGF在转染组中表达上调(P<0.05),导致NIH/3T3细胞增殖能力增强. 结论 EBV病毒miR BART5通过调控miR 18途径上调CTGF的表达,从而促进细胞的恶性表征.
