低氧微环境对胶质瘤细胞增殖和侵袭的影响

Abstract
目的:观察低氧微环境对胶质瘤细胞增殖和侵袭能力的影响,并进一步探讨其可能机制.方法:将胶质瘤细胞A172和U251分别置于普通细胞培养箱(常氧组:21%氧气含量)和低氧工作站(低氧组:2%氧气含量)中培养,采用CCK-8(Cell Counting Kit,CCK8)法检测细胞增殖能力,Transwell实验检测细胞侵袭能力,Western blot检测低氧诱导因子-1α(hypoxia inducible factor-1,HIF-1α)和Sox2表达水平.结果:低氧组胶质瘤细胞增殖和侵袭水平明显高于常氧组,且HIF-1α和Sox2表达水平也高于常氧组,差异具有统计学意义(P<0.05).结论:低氧微环境可能通过上调Sox2表达促进胶质瘤细胞A172和U251的增殖和侵袭.
