Abstract
目的: 观察和探讨CQMUH-011的抗炎作用及其机制.方法: 采用二甲苯致小鼠耳肿胀、大鼠棉球肉芽肿和佐剂诱导的大鼠类风湿性关节炎3种模型,观察CQMUH-011对不同炎症模型的抗炎作用,通过耳肿胀度、棉球肉芽肿干重、关节炎指数、足围肿胀度和踝关节病理切片反映相应炎症的严重程度,并利用酶联免疫吸附试验(enzyme-linked immunosorbent assay, ELISA)检测血清中肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)和白细胞介素6(interleukin-6, IL-6)的水平,相应的试剂盒检测一氧化氮(nitric oxide, NO)、丙二醛(malondialdehyde, MDA)和髓过氧化物酶(myeloperoxidase, MPO)水平.结果: 新化合物CQMUH-011能明显降低小鼠耳肿胀度、大鼠棉球肉芽肿干重和踝关节肿胀度,减轻关节炎指数,缓解关节病理损伤;并降低血清中TNF-α、IL-6和NO浓度,减少MDA含量,抑制MPO活性.结论: 新化合物CQMUH-011对炎症反应具有明显的抑制作用,这可能与其抑制炎症因子产生和抗脂质过氧化有关.
