Abstract
目的 构建小鼠Orai1基因的真核表达载体myc-Orai1(△4aa).方法 从小鼠脑组织中提取总RNA,RT-PCR扩增Orai1(△4aa)序列,所得片段及真核表达载体pRK5-myc用Sal I和Not I双酶切,后连入pRK5-myc载体中.重组质粒经Sal I和Not I双酶切鉴定后送公司测序.结果 PCR扩增得到预期900 bp大小的目的片段,经SalI和Not I双酶切鉴定正确重组质粒送公司测序,结果与预期一致.结论 成功构建myc-Orai1(△4aa)载体,为后续研究提供了条件.
