致倦库蚊卵黄羧肽酶基因与溴氰菊酯抗性关系的研究

Abstract
本文通过对不同抗性株致倦库蚊Culex pipiens quinquefasciatus卵黄羧肽酶基因(Vitellogeniccarboxypeptidase,VCP)表达量的比较,探讨VCP基因与溴氰菊酯抗药性的相关性.通过幼虫生物测定方法,分别得到致倦库蚊敏感株、野外株和抗性株,半数致死浓度分别为0.00057、0.38900和42.26250 mg/L.采用SYBR荧光定量方法,以致倦库蚊核糖体蛋白L8(ribosomal protein L8)基因作为内参,获得VCP基因在致倦库蚊敏感株、野外株和抗性株校正后(拷贝/l×109RPL8)表达量分别为5.80×10 7、4.53×107、3.07×10 7.致倦库蚊敏感株、野外株和抗性株的抗药性从低到高,而VCP基因表达量却是从高到低,推测两者之间存在着负相关关系.进一步推测VCP基因为致倦库蚊抗性相关的新基因,为研究致倦库蚊的抗药性提供了新的方向.
