过表达GINS2基因对NB4细胞增殖的影响

Abstract
目的 利用腺病毒载体Ad-GINS2感染NB4细胞,观察其对NB4细胞增殖的影响.方法 通过Ad-GINS2感染NB4细胞,利用荧光显微镜观察NB4细胞的感染情况,利用PCR和Western blotting技术检测GINS2基因的转录和翻译水平;MTT方法检测NB4细胞增殖情况.结果 PCR检测结果显示,与空载组和未处理组相比,GINS2的表达水平明显增加(P<0.05);Western blotting技术检测结果显示,与空载组和未处理组相比,GINS2的表达水平明显增加(P<0.05);MTT结果显示,与未处理组和空载组相比,重组腺病毒Ad-GINS2能明显促进NB4细胞的增殖(P<0.05).结论 Ad-GINS2成功感染NB4细胞并且促进NB4细胞增殖,为明确该基因在髓细胞白血病中的作用奠定了一定基础.
