DNA原位杂交法及斑点杂交法检测卡氏肺孢子虫的实验研究

Abstract
目的 探讨DNA原位杂交法及斑点杂交法在卡氏肺孢子虫检测中的应用.方法 设计合成卡氏肺孢子虫特异性寡核苷酸探针,建立卡氏肺孢子虫大鼠动物模型,分别于第6、8、10用处死动物,取肺组织提取DNA及石蜡切片进行斑点杂交及DNA原位杂交,结果同瑞氏-吉氏染色法比较.结果 DNA原位杂交法和斑点杂交法第6周检出阳性结果,检出率均为(30/30),高于瑞氏-吉氏染色法(25/30).结论 DNA原位杂交法及斑点杂交法是高效准确的卡氏肺孢子虫检测方法.
