广西人群miR-181 rs77418916和rs8108402多态性与系统性红斑狼疮的关联性研究

Abstract
目的 分析miR-181a rs77418916和miR-181c rs8108402单核苷酸多态性(SNP)与系统性红斑狼疮(SLE)患病的关联性.方法 用流式细胞术分析SLE患者淋巴细胞亚群,单碱基延伸PCR分型和DNA测序检测miR-181基因多态性,RT-qPCR检测miR-181a和miR-181c在单个核细胞中的表达水平.结果 rs8108402位点存在CC、CT和TT 3种基因型,CT和TT基因型及隐性和显性模型在SLE组与对照组比较差异均有统计学意义(CT vs CC:OR=1.50,95%CI:1.03~2.19,P=0.033;TT vs CC:OR=2.65,95%CI:1.18~5.98,P=0.019;CC/CT vs TT:OR=2.23,95%CI:1.01~4.93,P=0.048;TT/CT vs CC:OR=1.61,95%CI:1.12~2.31,P=0.010);rs77418916位点检测到AA、AT和TT 3种基因型,但未发现其与SLE遗传易感性相关;SLE组miR-181a和miR-181c基因的表达水平明显低于对照组(Z=-3.22,P<0.01;Z=-3.24,P<0.01),携带rs8108402 CT和TT基因型的患者miR-181c表达水平低于CC基因型携带者(Z=-2.51,P<0.05);SLE组CD3+、CD4+、CD8+和NK细胞绝对计数明显低于对照组(P<0.01).结论 miR-181c rs8108402多态性可能与广西人群SLE的遗传易感性相关,且CT或TT基因型携带者患SLE的风险增加,携带CT或TT基因型的个体可能通过抑制miR-181c基因表达从而增加SLE的发病风险.
