Abstract
Objective To study the effect of salvianolic acid A on expression of cytosolic phospholipase A2 (cPLA2) in rats following cerebral IR injury and its mechanism.Methods Sixty-five SD rats were randomly divided into control group,model group,salvianolic acid A treatment group and LY294002+salvianolic acid A treatment group (20 in each group).Brain tissue samples were taken from ischemic stroke rats at 0,8,24 and 48 h after reperfusion.Apoptosis was assayed by TUNEL,expressions of cPLA2,Akt and phosphorylated Akt were detected by Western blot withimmunohisto chemical staining.Results The number of TUNEL-positive cells was greater and the expression level of cPLA2 was significantly higher while the expression level of phosphorylated Akt was significantly lower in model group and LY294002-±-salvianolic acid A treatment group than in control group at 0,8,24,48 h after reperfusion (P<0.05).The expression level of phosphorylated Akt was significantly higher while the number of TUNEL-positive cells was significantly smaller and the expression level of cPLA2 was significantly lower in salvianolic acid A treatment group than in model group at 0,8,24,48 h after reperfusion (P<0.05).Conclusion Salvianolic acid A inhibits the expression of cPLA2 and reduces the apoptosis through the Akt pathway,thus playing a role in protecting cells in rats following IR injury.
