Abstract
目的 探讨慢病毒介导环状RNA mmu-circ-0001033过表达对低氧性小鼠肺动脉平滑肌细胞增殖的影响,为环状RNA防治低氧性肺动脉高压(HPH)提供实验依据.方法 体外原代培养小鼠肺动脉平滑肌细胞(PASMC),并随机分为4组:常氧组、低氧组、mmu-circ-0001033过表达慢病毒转染+低氧组、空白载体慢病毒转染+低氧组.体外构建环状RNA mmu-circ-0001033过表达慢病毒载体,转染PASMC.运用qRT-PCR技术检测各组PASMC中mmu-circ-0001033的表达水平,采用CCK-8法检测各组PASMC增殖,对各组的指标值进行统计学比较和分析.结果 低氧处理导致PASMC中mmu-circ-0001033的表达明显下调,且呈时间依赖性;与对照组比较,慢病毒转染组中mmu-circ-0001033表达显著上调(P<0.05),提示mmu-circ-0001033过表达慢病毒成功转染入PASMC,且并高效表达;与常氧组比较,低氧组和空白载体慢病毒转染+低氧组PASMC增殖明显增多(P<0.05),而mmu-circ-0001033过表达抑制了低氧对PASMC的促增殖作用.结论 过表达的mmu-circ-0001033显著抑制低氧所诱导的PASMC的增殖,为明确mmu-circ-0001033作为HPH防治靶标奠定了实验基础.
