Micro RNA-210诱导大鼠急性心肌梗死区域内微血管新生作用机制研究

Abstract
目的 研究micro RNA-210(miRNA-210)在大鼠急性心肌梗死(AMI)中调节微血管新生作用并探讨其相关机制.方法 将大鼠按1:1随机分为:假手术组(Sham组)、心肌梗死+阴性病毒对照组(AMI+NV)、心肌梗死组(AMI+PBS)、心肌梗死+miRNA-210激动剂体内转染组(AMI+miRNA-210 agonist).术后4周以微血管密度(MVD)为效应终点,检测各组内miRNA-210、肝细胞生长因子(HGF)、血管内皮生长因子(VEGF)的mRNA;检测HGF及肌球蛋白重链(β-MHC)的表达,小干扰RNA(siRNA)处理HGF,以WB检测效应终点.结果 AMI+miRNA-210 agonist组HGF表达水平、微血管密度(MVD)随miRNA-210表达增加而显著增加;低氧环境下siRNA显著抑制HUVECs中HGF表达,CD 31表达水平也显著下降;AMI+miRNA-210 agonist组大鼠心脏收缩功能更为优越.结论 心肌内miRNA-210过表达可上调梗死区域内HGF表达水平,促进血管新生,改善心脏收缩功能.
