Abstract
BCR/ABL融合蛋白通过改变酪氨酸磷酸化水平,进而活化一系列信号转导路径,在慢性髓系白血病(chronic myeloid leukemia,CML)发病中发挥关键作用.酪氨酸激酶抑制剂(tyrosine kinase inhibitors,TKIs)通过竞争性抑制ATP与催化中心的结合,阻断酪氨酸激酶活化,达到治疗目的.以伊马替尼为代表的TKIs开启了CML分子靶向治疗的新时代,已取代干扰素-α和干细胞移植作为一线治疗,目前CML治疗10年总生存率达85％～90％.
