microRNA let －7f －5p 对胃癌细胞系 GC9811迁移和侵袭能力的影响

目的：探讨转染 let －7f －5p mimics／inhibitor 对胃癌细胞系 GC9811迁移和侵袭能力的影响。方法：用实时定量聚合酶链反应（RT －PCR）的方法在 mRNA 水平测定转染 let －7f －5p mimics／inhibitor 及其阴性对照后 let －7f －5p 表达水平的变化；以 Transwell 实验检测转染 let －7f －5p mimics／inhibitor 后迁移和侵袭能力有无变化。结果：RT －PCR 结果显示：转染 let －7f －5p mimics 可明显上调 GC9811细胞 let －7f －5p 的表达；转染 let －7f －5p inhibitor 可明显下调 GC9811细胞 let －7f －5p 的表达。Transwell 实验结果显示与对照组相比转染 let －7f －5p mimics 的 GC9811细胞迁移和侵袭能力明显减弱；与对照组相比转染 let －7f －5p inhibitor的 GC9811细胞迁移和侵袭能力明显增强。结论：microRNA let －7f －5p 能够抑制胃癌细胞 GC9811的迁移和侵袭能力。
