Abstract
目的:探讨Tim-4促进子宫颈癌细胞侵袭与迁移及其相关机制.方法:用实时荧光定量PCR法检测子宫颈癌细胞系Siha、Hela与子宫颈上皮永生化细胞系H8中Tim-4的表达水平.在较低表达Tim-4的Siha细胞系感染携Tim-4的慢病毒载体,荧光显微镜观测Siha细胞系转染Tim-4的表达水平.用Transwell及划痕实验检测Tim-4对子宫颈癌细胞系Siha的侵袭与迁转移能力的影响,用Western blotting检测Siha细胞转染Tim-4后MMP2、MMP9、E-cadherin、N-cadherin的变化.结果:子宫颈癌细胞系Siha、Hela与宫颈上皮永生化细胞系H8相比高表达Tim-4.成功构建携Tim-4慢病毒载体的Siha细胞系;Tim-4明显抑制子宫颈癌细胞系Siha的迁移与侵袭能力,且影响了MMP2、MMP9及N-cadherin、E-cadherin的表达水平.结论:过表达Tim-4可促进子宫颈癌细胞的侵袭与迁移能力,其机制可能与Tim-4促进Siha细胞EMT转化有关.
