碱性成纤维细胞生长因子对3D培养条件下牙髓干细胞成牙分化的研究

Abstract
目的:探讨碱性成纤维细胞生长因子(basic fibroblast growth factor, FGF-2)对3D培养的牙髓干细胞(dental pulp stem cells,DPSCs)成牙分化的影响.方法:体外分离和培养DPSCs,通过成牙/成骨和成脂诱导,鉴定其多向分化的潜能;构建纳米纤维微球,将 DPSCs 与纳米纤维微球复合3D 培养,FGF -2 (20 ng/mL)刺激DPSCs 7 d,常规培养7 d,诱导培养7 d后,实时定量PCR和免疫荧光染色分别测定DPSCs成牙分化相关基因和蛋白的表达变化.结果:体外成功培养的DPSCs,具有成牙/成骨和成脂方向分化的潜能;FGF-2刺激3D培养的DPSCs后,成牙分化相关基因ALP、DMP1、OCN、DSPP以及成牙分化相关蛋白在2周时显著升高(P<0.05).结论:FGF-2可效促进DPSCs向成牙本质样细胞分化.
