Abstract
目的:建立UPLC-MS/MS法测定猴头菌丝体中18种游离氨基酸的含量.方法:采用Hypersil GOLDC18色谱柱(100 mm×2.1 mm,1.9 μm),以乙腈-0.1％甲酸溶液为流动相,梯度洗脱,流速0.2 mL· min-1;以电喷雾离子源(ESI)正离子模式将样品离子化,多反应监测(MRM)模式下对18种氨基酸进行测定.结果:18种氨基酸相关系数为0.999 3～1.000;平均回收率(n=6)为98.3％～100.0％,RSD在0.34％～1.5％.9批猴头菌丝体样品中,18种游离氨基酸总量在72.1～97.3 mg· g-1之间.结论:本法可作为猴头菌丝体中游离氨基酸的含量测定方法.
