Abstract
目的:建立了HPLC-MS/MS法测定人体血浆中人参皂苷Rh2的浓度,并用于人参次苷H滴丸的药动学研究.方法:采用Welch Materials Column XB-C18色谱柱(100 mm×4.6 mm,5μm),以甲醇-水为流动相,流速为0.4 mL· min-,柱温40℃.以阿立哌唑为内标,血浆样本碱化后,以二氯甲烷和乙醚(3∶2,v∶v)萃取,40℃水浴挥干后用流动相复溶进样.采用多反应离子监测,正离子模式扫描,电喷雾离子源.结果:建立的HPLC-MS/MS法在0.5 ~200 ng·mL-1范围内人参皂苷Rh2色谱响应与浓度相关性良好,定量下限为0.5ng·mL-1;人参皂苷Rh2及内标的保留时间分别为14.10和6.61 min;准确度、精密度、回收率和经内标校准后的基质效应均合格;稳定性各项数据均符合相关要求.结论:该法灵敏、专属性强、准确性好,适用于人血浆中人参皂苷Rh2的浓度测定.
