高表达Per2基因的胶质瘤干细胞下调其基质 金属蛋白酶2的研究

Abstract
目的 构建Period2(Per2)基因过表达的U87胶质瘤干细胞,检测Per2基因过表达U87胶质瘤干细胞中基质金属蛋白酶2(MMP-2)的mRNA及蛋白的表达变化.方法 以Per2基因过表达慢病毒转染U87胶质瘤干细胞,然后对U87胶质瘤干细胞进行转染,通过嘌呤霉素筛选,用real-time PCR和Western blot技术鉴定得到Per2基因过表达的U87胶质瘤干细胞,空载体转染的U87胶质瘤干细胞和U87胶质瘤干细胞未转染组作为对照.结果 此次转染成功构建Per2基因过表达U87胶质瘤干细胞系,过表达组细胞内Per2基因的mRNA和蛋白表达量明显高于其他两组细胞,MMP-2 mRNA及蛋白表达量在Per2基因过表达U87胶质瘤干细胞内明显降低.结论 U87胶质瘤干细胞高表达Per2基因能够下调其MMP-2.
