沙格雷酯下调肺动脉高压大鼠右室TRPC1、TRPC6通道及calcineurin A/NFATc3表达

Abstract
目的 研究5-HT2A受体拮抗剂沙格雷酯对肺动脉高压大鼠的右室TRPC1,TRPC6通道及calcineurin A/NFATc3表达的影响. 方法 18只大鼠分为对照组、肺动脉高压组和沙格雷酯组,每组6只.对照组给予生理盐水腹腔注射和生理盐水灌胃,肺动脉高压组给予野百合碱腹腔注射及生理盐水灌胃,沙格雷酯组给予野百合碱腹腔注射及沙格雷酯灌胃.干预4周时采用RT-PCR和Western blot检测右室TRPC1、TRPC6通道和calcineurin A/NFATc3的mRNA和蛋白表达,并对三组间结果进行比较. 结果 与对照组相比,肺动脉高压组大鼠右室心肌中TRPC1、TRPC6通道、calcineurin A/NFATc3的mRNA和蛋白表达升高(P<0.05);与肺动脉高压组相比,沙格雷酯组大鼠右室心肌TRPC6通道、calcineurin A/NFATc3的mRNA表达降低,TRPC1、TRPC6通道、calcineurin A/NFATc3的蛋白表达降低(P<0.05). 结论 肺动脉高压大鼠右室心肌的TRPC1、TRPC6通道、calcineurin A/NFATc3表达上调,沙格雷酯可逆转这些蛋白和基因的表达水平.
