基因Protocadherin-PC对前列腺癌细胞上皮间质化生的影响

目的 为了分析基因Protocadherin-PC与前列腺癌转移是否有关,研究其对前列腺癌细胞上皮间质化生的影响,从而探索基因Protocadherin-PC与前列腺癌细胞侵袭转移的关系.方法 分别采用蛋白免疫印记、形态检测、细胞划痕闭合实验的方法检测通过Protocadherin-PC siRNA抑制基因Protocadherin-PC表达对雄激素非依赖性前列腺癌细胞DU-145细胞、PC-3细胞上皮间质化生的影响.结果 蛋白免疫印记结果提示抑制基因Protocad-herin-PC表达可以促进DU-145细胞、PC-3细胞间质上皮化生;形态学检测提示抑制基因Protocadherin-PC表达在形态上可以诱导DU-145细胞、PC-3细胞更接近于雄激素依赖性前列腺癌细胞LNCaP;细胞划痕闭合实验结果提示抑制基因Protocadherin-PC表达可以减慢DU-145细胞、PC-3细胞生长速度.结论 通过Protocadherin-PC siRNA抑制基因Protocadherin-PC表达可以降低雄激素非依赖性前列腺癌细胞DU-145、PC-3侵袭转移力,从而揭示基因Protocadherin-PC又一新功能,与晚期前列腺癌转移关系密切.
