Abstract
目的 利用PKH26和Hoechst 33258联合示踪Uncv无毛小鼠iRhom2及其突变体蛋白在Vero细胞中的定位.方法 用PKH26对细胞膜进行染色,同时用Hoechst 33258染料对细胞核进行染色,置于激光共聚焦显微镜下观察.结果 通过激光共聚焦荧光显微镜观察目的蛋白,发现野生型iRhom2分布在细胞的胞浆,而iRhom2mut于细胞浆内和细胞核内都存在.结论 亚细胞定位的不同推测到iRhom2的N末端缺失突变可能对其细胞内的定位有影响.
