白芨多糖抑制HepG2细胞增殖研究

Abstract
目的:研究白芨多糖对HepG2增殖的影响.方法:在HepG2培养基中分别加入终浓度为0、0.5mg/mL、1.0mg/mL、1.5mg/mL的白芨多糖,观察细胞贴壁和形态的变化,采用MTT检测细胞的增殖情况,分别于0、24h、48h对1.0mg/mL白芨多糖处理的细胞进行基因组电泳和caspase-3 RT-PCR检测,研究白芨多糖是否可以诱导HepG2凋亡.结果:1.0mg/mL以上浓度的白芨多糖对HepG2贴壁有抑制作用,0～1.5 mg/mL白芨多糖不抑制HepG2的分裂,1.0mg/mL的白芨多糖可诱导HepG2细胞凋亡.结论:1.0mg/mL的白芨多糖具有抑制HepG2细胞增殖的作用,作用机理可能为抑制细胞贴壁和诱导HepG2细胞凋亡.
