低能N+离子注入诱变选育金耳多糖高产菌株的研究

Abstract
应用低能N+离子注入金耳酵母状分生孢子菌株进行诱变,以获得一株多糖产量更高的金耳诱变菌株.试验确定了低能N+离子注入的适宜参数:注入剂量为60× 1014 ions/cm2,注入能量20 KeV,靶室真空度10-3 Pa.以5s脉冲式注入,间隔时间为15s.离子注入后,分别以液体发酵生物量(孢子量)及多糖产量为初筛和复筛标准,经遗传稳定性试验验证,筛选到了一株金耳多糖高产菌株,其产量较对照菌株提高了12.3%,液体发酵多糖产量(包括菌体多糖与发酵液多糖)达2.7 g/L.结果 表明:低能N+离子注入是一种较为理想的金耳诱变育种手段.
