响应面法优化芽孢杆菌NBIN-863的发酵工艺

Abstract
采用响应面分析法对苏云金芽孢杆菌(Bacillus thuringiensis)NBIN-863的发酵培养基进行优化,应用Plackett-Burman设计法对影响NBIN-863发酵的关键因子进行筛选,通过最陡爬坡路径法确定主要影响因素的响应中心点,并进一步采用中心组合设计和响应面分析法确定最优培养基.将发酵液稀释后进行线虫毒性研究,结果表明,影响该菌株芽孢产量的关键因素为淀粉和豆粕的浓度.通过响应面分析法的拟合和推算,得到淀粉和豆粕浓度分别为35.2 g/L和34.7 g/L,测得NBIN-863发酵液的晶体含量为5.4 mg/mL,稀释后对线虫的致死率为81.0%,与预测值(81.2%)十分接近,比优化前NBIN-863的实际致死率(45.0%)提高了80%.
