酒鬼酒发酵车间空气源细菌产酯酶活性菌株筛选及多样性分析

Abstract
采用平板水解圈法(plate hydrolysis spot method)对分离自酒鬼酒发酵车间空气的细菌进行产酯酶活性菌株筛选,采用基于16S rRNA基因序列的分析方法对高酶活菌株进行系统发育多样性分析。结果表明,73个受试菌株中,有51株酯水解酶活性菌株呈阳性,占受试菌株的69.9%。其中有9株(JSM 2215018、JSM 2215021、JSM 2215027、JSM 2215104、JSM 2215109、JSM 2215116、JSM 2215117、JSM 2215126、JSM 2215132)具有较强的酯酶活性。系统发育分析表明,这9株菌属于细菌域(Eubacteria)的4个大的系统发育类群(Actinobacteria, Deinococcus-Thermus, Firmicutes, Proteobacteria)的8个科(Bacillaceae, Deinococcaceae, Microbacteriaceae, Micrococcaceae, Mo-raxellaceae, Nocardiaceae, Rhodobacteraceae, Xanthomonadaceae)的8个属:JSM 2215018属于Microbacterium属;JSM 2215021属于Arthrobacter属;JSM 2215027属于Bacillus属;JSM 2215104属于Xanthomonas属;JSM 2215109属于Deinococcus属;JSM 2215116属于Acinetobacter属;JSM 2215117和JSM 2215132属于Paracoccus属;JSM 2215126属于Rhodococcus属。研究结果表明,酒鬼酒发酵车间空气源细菌存在较高比例的产酯酶菌株,且这些菌株具有较高的类群多样性。
