金花茶通过AMPK/PPAR-α通路减轻NAFLD引起的脂代谢异常机制研究

Abstract
[目的]观察金花茶(Camellia chrysantha(Hu) Tuyama)对非酒精性脂肪性肝病(non-alcoholic fatty liver disease,NAFLD)的防治效果,初步研究其作用机制.[方法]将48只C57BL/6小鼠随机分为6组:对照组、NAFLD模型组、金花茶粉末低剂量组、金花茶粉末高剂量组、金花茶水提物低剂量组、金花茶水提物高剂量组,每组各8只.除对照组采用普通饲料喂养外,其他各组采用高脂膳食喂养8周建立NAFLD模型,喂养结束后检测小鼠的体重、脂肪重量、血清生化指标、肝组织中极低密度脂蛋白受体(very low density lipoprotein receptor,VLDLR)、过氧化物酶体增殖物激活受体-α(peroxisome proliferator activated receptor-α,PPAR-α)与磷酸化腺苷酸激活蛋白激酶(phosphorylated-adenosine monophosphate activated protein kinase,p-AMPK)蛋白的表达水平变化.[结果]低剂量金花荼粉末与水提物均可以显著降低NAFLD小鼠血清中甘油三酯(triglyceride,TG)、谷丙转氨酶(alanine aminotransferase,ALT)和谷草转氨酶(aspartate transaminase,AST)水平(P<0.05),其中低剂量金花茶水提物显著降低NAFLD小鼠肝组织中VLDLR表达水平(P<0.05),显著提高p-AMPK与PPAR-α蛋白表达水平(P<0.05).[结论]金花茶水提物通过下调VLDLR蛋白表达,上调p-AMPK、PPAR-α蛋白表达,减轻NAFLD引起的脂代谢异常,缓解NAFLD小鼠肝脏脂质病变.
